Red Wine Molecule Resveratrol Promotes A Type Of T-Cell That Literally Aborts Infection And Symptoms Typically Seen In These Workers.
A remarkable discovery….. healthcare workers, who by repeated exposure to corona cold viruses in their line of work, have pre-existing T-cell immunity against prior coronavirus infections that confer immunity against COVID-19 to the point of aborting infection BEFORE antibodies can even be generated (even before a swab or blood test can detect it!). These subjects would then test NEGATIVE for COVID-19, having already aborted the infection and consequent symptoms. This startling discovery is documented in a landmark report published in Nature Magazine.
Initially the immune system is alerted to a new foreign virus and responds with an array of white blood cells (innate immune response), namely neutrophils and macrophages, to battle the virus in a non-specific way, while 5-7 days later T-cells and B-cells develop specific memory antibodies and memory T-cells that provide lasting immunity (adaptive immune response).
But in this instance, pre-developed immunity against prior coronavirus infections (MERS*, SARS* or other corona cold viruses) produced cross-immunity against COVID-19. With prior pathogenic coronavirus strains sharing 82% similarity to gene sequences in COVID-19, researchers say the re-purposing of existing natural inhibitors of coronaviruses is in order.
Replication-Transcription Complex Enzymes & Resveratrol
Of specific interest are T-cells produced by replication-transcription complex (RTC) enzymes. RTC enzymes are what drive the molecular machinery of RNA synthesis, COVID-19 being an RNA virus.
Healthcare workers with the stronger RTC-specific T-cells exhibit “robust early innate immunity” to the point of aborting the infection altogether!
The largest and most essential of these RTC enzymes in coronaviruses is Nsp3 (non-structural protein 3). The red wine molecule resveratrol is on a list of natural molecules that inhibit Nsp3.
Structural proteins in COVID-19
As researchers reveal, there are four structural proteins within coronaviruses: M (membrane); S (spike); E (envelope) and N (neocapsids), essential for production of complete infective forms of viruses outside a host cell called virions, housed within a protein shell called a capsid.
The spike (S), membrane (M), envelope (E) proteins combine within a capsid (N) to form a mature virion that is then released from the host cell to infect other cells.
The N protein (enzyme) is the only structural protein associated with RTCs (replication-transcription complex), the critical component found in healthcare workers that conferred such powerful immunity it aborted infection and symptoms altogether.
The N protein is an important target for biologists to develop therapies that block viral replication within infected cells, probably more important than spike protein that the current RNA vaccines are fashioned around.
Resveratrol targets N protein, essential for viral replication
While researchers say this type of immunity can be accomplished with small molecules that can enter genetic machinery and they beg for research to begin, the library of natural molecules already offers such a molecule.
Of considerable interest is that resveratrol, a natural molecule derived from grapes and wine, targets and inhibits the N protein that is essential for viral replication and thus prolongs cellular survival even after viral infection!
This was demonstrated by direct instillation of resveratrol in a lab dish two years prior to the COVID-19 pandemic! Researchers stated: “Resveratrol remarkably inhibits coronavirus nucleocapsid N-protein translation.” While this was only demonstrated at high-dose resveratrol concentration, repeat administration of resveratrol lowered the dose required to produce a virus-killing effect.
DPP4: Another target to block COVID-19 coronavirus
Another enzyme called DPP4 (dipeptidyl peptidase) is a major receptor (entry point) for coronaviruses and would also serve as a test target for resveratrol. In fact, resveratrol has been demonstrated to inhibit DPP4, an enzyme also involved in blood sugar control. Diabetics are at risk for more severe Covid-19 infection.
Experts comment about this discovery
Professor Peter Openshaw, professor of Experimental Medicine at Imperial College in London, says: RTC-specific T-cells “abort infection before symptoms develop, presumably by acting within the mucosa… Vaccines designed to induce RTC-specific T-cells might have broad protective effects against endemic and emerging coronaviruses and might prevent disease from occurring.”
Dr. Andrew Freedman, Reader in Infectious Diseases and Honorary Consultant Physician, Cardiff University School of Medicine, said: “The immune system was able to clear the virus rapidly before it could cause symptoms or be detected either by PCR test or antibody production.”
Dr Alexander Edwards, Associate Professor in Biomedical Technology, University of Reading, said these healthcare workers had “enough virus exposure to activate part of their immune system, but not enough to experience symptoms, detect significant levels of virus, or mount an antibody response.”
Edwards went on to say: “A vaccine that primes T-cell immunity against different viral protein targets that are shared between many different coronaviruses would complement spike protein vaccines that induce neutralizing antibodies.” Would resveratrol be that “vaccine?”
